I play audio from this DFplayer mini mp3 player https://www.dfrobot.com/wiki/index.php/DFPlayer_Mini_SKU:DFR0299
I have connected this player to the 50W x 2+100W 2.1 Channel subwoofer amplifier https://www.amazon.com/gp/product/B01NACU4PW/ref=ppx_yo_dt_b_asin_title_o06_s00?ie=UTF8&psc=1
and the amplifier is connected to 4 ohm 40Watts car speaker and everything works fine.

But whenever I substitute the speaker with this horn speaker https://www.amazon.com/gp/product/B077YJK4VG/ref=ppx_yo_dt_b_asin_title_o06_s00?ie=UTF8&psc=1 It doesn't work, it sounds like the audio wants to come on but it is chocked or something kind of breaking up and very low. This speaker supposedly has built-in amplifier and I have also have tried another horn speaker without built in amplifier but it was the same outcome.I am very new to audio stuff, what am I doing wrong here.

Thank you in advance.

There's no way to connect the power horn speaker to an amp.
How were you trying to hook it up?

I tried to hook up horn speaker red and black to the Amp output, I kinda hear the audio but it's breaking up and very low.If that is not possible how can I use the horn speaker with this mp3 player in this case?

Just making sure if I understood you, so there is no way at all to connect horn speakers(even the ones without built-in Amp) to an Amp?
Not exactly. A horn speaker without an amp is just a speaker in my book.
No you cannot connect the power horn to the 2.1 channel amp output you linked in post #1
You mentioned you tried a horn speaker without an amp and it did not work. What horn speaker was that?

It may be possible to connect the output from the MP3 player to the power horn.
From the photos of the power horn there is a 4 conductor female receptacle using 3 wires, red, green and yellow. Most likely those wires are power, ground and audio out from the microphone. If you can figure out these wires you can try connecting the DAC_ L and DAC_ R outputs to the ground and audio out of said receptacle.

I agree. Those wires you are using are the power wires to the amplifier built into the back of the horn. If you want to use the horn speaker on your class D amp, first you will have to pull the back off the horn and disconnect the actual horn speaker from the internal amp.
But why do you want to do that in the first place? If you try to run the horn on a bigger amp that it is designed for you have a very great risk of killing it. Buy a new horn speaker with the appropriate power rating for the amp in question.
