May I know what are the parameters we will tune when we face setup and hold time issues in high speed PCB.

Match line lengths, insert delays, add latches and use multi-cycle paths, ...

 

But most important pay attention to the setup and hold time requirements of the devices you are planning to use on the board and incorporate the transmission line (or other) delays in your calculations.

May I know how to insert transmission line delay.

One way is to insert additional trace length. If you need a large delay, you might insert a buffer.
