Lets ask some very basics question.

What is the function of VR ?

My question - What will happen if LED anode voltage becomes higher than 5V lets say 6V max.
Since reverse voltage of LED is VR = 5V, will led get damage ?

In your circuit there will never be any reverse voltage. Don't worry about it.

If you put a 6V power supply across the diode backwards, so no light, then the diode might get hot and die.

 

And if you put 6V across in the forward direction, you will get a brilliant flash.

I assume when LED is off only LED anode see 6V since 6V is dc source not AC.

So no damage at all.

 

What is the function of VR ?

It is a maximum reverse voltage rating.
If you exceed that reverse voltage it may damage the LED.
