please tell us what voltage we should we apply to diode 1N4001 for it's reverse (leakage) current test?
and how can we confirm that the diode is ok or not after result.
same way also tell for forward current test of diode?

Well, what voltage does the data sheet give as the peak reverse voltage?

You need to be able to apply (and measure) the voltage across the diode as well as the current through the diode. The data sheet will give the scale of the voltages and currents involved for each test. You want to be able to limit the current so as not to exceed the specified limits. You also need to consider that your test circuit has to be able to measure the voltages and currents accurately enough for your purposes -- which if you are within a few percent that is probably good enough.
