Is is possible to replace the coil by a real single-end transfo
like Hammond 125DSE ?

If you put the single ended transformer on a Network Analyzer and it sweeps the same s-parameters as the original coil then I'd say yes go ahead and make the substitution. You did not say what kind of a tube amplifier this is so I'm kinda guessing here.
