I want to send one serial command to multiple chips rather than using the secondary UART's on the 2560 to control the other's (easier from a programming stand point). Can the FTDI FT232RL support multiple devices connected to its TX line (Pin 1)?

Provided you don't exceed the fanout, I don't see a reason it wouldn't be able to.

Careful, you asked about multiple devices listening, nothing about a response from the listeners...

Are you planning to only have one device physically connected to Rx on the FT232?

 

All 2560's will receive data via the TX line, but only one 2560 will be connected back to the RX Line of the FT232.
